چکیده
Abstract Background Mitochondrial dysfunction is a critical factor in the onset and progression of neurodegenerative diseases. Recently, mitochondrial transplantation has been advised as an innovative attractive strategy to transfer replace damaged mitochondria. Here we propose, for first time, use rat brain extracted synaptosomes, subcellular fraction isolated synaptic terminal that contains mitochondria, delivery systems. Results Synaptosome preparation was validated by presence Synaptophysin PSD95. Synaptosomes were characterized terms dimension, zeta potential, polydispersity index number particles/ml. Nile Red or CTX-FITCH labeled synaptosomes internalized LAN5 recipient cells mechanism involving specific protein–protein interaction, demonstrated loss fusion ability after trypsin treatment using different cell lines. The loading release proved curcumin both into cells. vitality mitochondria transferred Opa1, Fis1 TOM40 proteins JC-1 measurements. Further, deliver vital cytoplasm neuronal microscopic images, increase TOM 40, cytochrome c, Hexokinase II proteins, DNA. Finally, vehicle, healthy restored function containing rotenone CCCp Conclusions Taken together these results suggest can be natural vehicle molecules organelles replacement affected with ones could potential therapy treating dysfunction-related

A new pathway for mitochondrial quality control: mitochondrial-derived vesicles.
The last decade has been marked by tremendous progress in our understanding of the cell biology of mitochondria, with the identification of molecules and mechanisms that regulate their fusion, fission, motility, and the architectural transitions within the inner membrane. More importantly, the manipulation of these machineries in tissues has provided links between mitochondrial dynamics and phy...
متن کاملSynaptosomes as a platform for loading nanoparticles into synaptic vesicles.
Synaptosomes are intact, isolated nerve terminals that contain the necessary machinery to recycle synaptic vesicles via endocytosis and exocytosis upon stimulation. Here we use this property of synaptosomes to load quantum dots into synaptic vesicles. Vesicles are then isolated from the synaptosomes, providing a method to probe isolated, individual synaptic vesicles where each vesicle contains ...
متن کاملPalmitoyl protein thioesterase (PPT) localizes into synaptosomes and synaptic vesicles in neurons: implications for infantile neuronal ceroid lipofuscinosis (INCL).
A deficiency of palmitoyl protein thioesterase (PPT) leads to the neurodegenerative disease infantile neuronal ceroid lipofuscinosis (INCL), which is characterized by an almost complete loss of cortical neurons. PPT expressed in COS-1 cells is recognized by the mannose-6-phosphate receptor (M6PR) and is routed to lysosome, but a substantial fraction of PPT is secreted. We have here determined t...
متن کاملDecylubiquinone increases mitochondrial function in synaptosomes.
The effects of decylubiquinone, a ubiquinone analogue, on mitochondrial function and inhibition thresholds of the electron transport chain enzyme complexes in synaptosomes were investigated. Decylubiquinone increased complex I/III and complex II/III activities by 64 and 80%, respectively, and attenuated reductions in oxygen consumption at high concentrations of the complex III inhibitor myxothi...
متن کاملThe separation of synaptic vesicles from nerve-ending particles ('synaptosomes').
When brain tissue is homogenized in media iso-osmotic to plasma, the club-like presynaptic nerve endings resist disruption and are snapped or torn off from their attachments to form discrete particles (nerve-ending particles) in which all the main structural features of the nerve ending are preserved. For these particles we propose the name ' synaptosomes' in order to emphasize their relative h...
